My college thesis was completed with Prof. Eric Wieschaus (Nobel ‘95) at Princeton University, where I studied Drosophila fruit fly embryogenesis. We examined the bicoid RNA-binding protein and its morphogen gradient noise, precision and expression dynamics. After graduating summa cum laude, I joined GIS for a year as a National Science Scholar, under the guidance of Dr. Lim Bing, to study human lung cancer stem cells. In collaboration with Dr. Harvey Lodish at MIT, we also examined how vertebrate lin-4 (or miR-125b) regulates stem cell self-renewal and neuroblastoma – the most common cancer in infants. At Harvard Medical School, I trained under the mentorship of Prof. George Daley at Boston Children’s Hospital. We focused on the RNA binding protein lin-28, and its targets the pro-differentiation let-7 microRNAs, and how they coordinate glucose metabolism and insulin-PI3K signaling in embryonic stem cells and skeletal muscle myoblasts during aging. Lin28a/b reactivation in muscles prevents obesity and diabetes, whereas let-7 microRNAs (which accumulate with aging) promote insulin resistance and type 2 diabetes. Some of our statistical calculations were completed in collaboration with David Altshuler, then at Broad Institute. By reactivating Lin28a, which declines during normal development and aging, in multiple adult tissues, we could also partially restore fetal-like tissue repair and wound healing capacities in adult mammals. Lin28a accomplishes this by reactivating an embryonic state of mitochondrial bioenergetic metabolism. This contributes to solving a mystery long discussed by Charles Darwin and others, of how youthfulness is often correlated with better tissue regeneration across the animal kingdom. Then, under the mentorship of Prof. Lewis Cantley, I used an unbiased LC-MS/MS metabolomics approach to elucidate the connections between nutrition and stem cell fate. We discovered how mitochondrial amino acid (Thr-Gly-Met) metabolism controls histone methylation during iPS reprogramming of somatic fibroblasts into pluripotent stem cells. This discovery connects nutritional metabolism to the epigenetic state of stem cells, with important clinical ramifications for cancer cells since their Gly metabolism is also often dysregulated. At the IOZ CAS, and previously at GIS A*STAR, my lab is using a variety of animal models to investigate stem cell metabolism during embryogenesis, tissue regeneration, and aging-induced degeneration, particularly in skeletal muscles. My vision is to use the latest technologies in RNA-sequencing, mass spectrometry, and 3D organoid cultures, to understand how we can modulate metabolic networks to control muscle stem cell (MuSC) self-renewal, differentiation, and aging. This will have important implications for treating degenerative diseases such as type 2 diabetes, sarcopenic obesity, and cancer cachexia, during human aging. show More
